- Most Probable Number Method 8368, A-1 Medium, is USEPA-accepted for testing non-potable waters. Method 8368 meets or exceeds the specification criteria stated in Standard Methods for the Examination of Water and Wastewater, 18th edition, 9221 E. Fecal Coliform Procedure. USEPA Manual for the Certification of Laboratories Analyzing Drinking Water states “5.5.3. A-1 medium may be used as an alternative to EC Medium to enumerate fecal coliforms in source water, in accordance with the Surface Water Treatment Rule. A-1 Medium must not be used for drinking water samples.”

Media Decontamination
- Recommendation:
- Open each MPN tube and pipet in 2-3 mls of Clorox. (you can use a disposable plastic droppers).
- Cap and invert …. Let stand for minimum 20-30 minutes.
- After waiting time, use a wire strainer or colander, empty the contents of the MPN tubes in to the strainer with the TAP water running. Rinse the tubes with TAP water.
- The empty tubes can now be disposed of in the glass waste container. Caps can be disposed of in the regular waste can.
- After all the tubes have been dosed, add additional amount of Clorox down the drain, ~20 mls. Run more Tap water into the sink.
