The Oxygen Demand, Chemical-Reactor Digestion Method 8000 does use 2mL of DI water to create the reagent blank, and that vial should be stored in the dark and re-used. The 5mL in step 2 (page 4) is added to an empty vial and is used to set the instrument zero (step 3) before measuring and recording the absorbance of the reagent blank vial discussed above (step 4), this provides a reproducible reference point for monitoring changes to the absorbance of the blank.
