Answer
Extreme pH can effect the color formation of the PhosVer® 3 phosphate reagent in methods 8048 and 8190. The sample should be near pH 7 prior to adding the PhosVer® 3 phosphate reagent. If samples have been acidified from digestion for total phosphorus determination, 5.0 N sodium hydroxide should be used to adjust to pH 7. After the addition of the PhosVer® 3 phosphate reagent, the sample should be pH 1-2.
